Properties

Over time, inflammation becomes frequent. In addition to those that become chronic, there are also the alterations of everyday life. Joints become less resistant with age, as cartilage is less able to repair itself, stimulating self-perpetuating inflammation. To get out of this situation, you have to go back to the original mediator. Joint inflammation is characterized by the Latin terms rubor, dolor, calor:

  • Persistent acute or chronic pain,
  • Swelling, a swollen joint, warm to the touch and red from the accumulation of synovial fluid,
  • Morning stiffness of joints,
  • Loss of mobility

Inflammation of the tissues (joints) is regulated by a number of molecules, the implementation of which is controlled by a single gene, NF-κB. This gene encodes the transcription factor that activates the synthesis of cytokines, prostaglandins and leukotrienes. When a joint or tissue is damaged or infected, the immune system releases these substances to initiate the first stage of repair (healing). Sometimes, particularly in the case of autoimmune diseases, the immune system mistakenly attacks tissues, causing chronic inflammation. The transcription factor NF-κB is the initial mediator of all inflammatory responses.

In vitro tests carried out by Laboratoires TEXINFINE on combined extracts of Castanea sativa and Portulaca oleracea showed inhibition by over 50% of the transcription factor NF-κB encoded by the NF-κB gene, and of the two cytokines (IL-1β, PGE2) which play a major role in inflammatory processes.

Vitamin K2 contributes to joint suppleness. TEXINFINE has chosen MenaQ7® Vitamin K2, derived from the fermentation of natural sources, as the source of vitamin K2 for FAGAFLEX®. This natural vitamin K2 in the form of menaquinone-7 (MK-7) is the most bioavailable and bioactive form of vitamin K in inflammation. MenaQ7® Vitamin K2 has been clinically tested in the treatment of rheumatoid arthritis, where menaquinones (MK) induce apoptosis of synovial cells in rheumatoid arthritis (RA).

Vitamin C ensures proper synthesis of collagen, a protein essential to cartilage structure, enabling optimal joint function.

Synoviocytes are specialized fibroblasts located in the synovial membranes. Their main role is the production of joint lubricants. Under pathological conditions (arthritis), these cells can acquire an aggressive phenotype characterized by the production of inflammatory cytokines and catabolic (destructive) enzymes. This phenotypic modulation has long been considered characteristic of inflammatory forms of arthritis such as rheumatoid arthritis. More recent studies (Mathiessen and Conaghan, 2017) have highlighted that this phenotypic transformation can lead to osteoarthritis with cartilage degeneration. Indeed, synoviocytes are capable of producing proinflammatory mediators (IL-1, IL-6, TNF-α, PGE2, etc.) stimulating the expression of catabolic enzymes (such as metalloproteinases also known as MMP-1, 3, 9, 13) responsible for osteoarticular degradation. The use of plant extracts with inhibitory activity on the expression of pro-inflammatory molecules by synoviocytes may offer effective therapeutic potential against deterioration in joint integrity, particularly in conditions of chronic inflammation linked to pathologies (osteoarthritis) or aging.

There are two forms of vitamin K: vitamin K1 (phylloquinone) and vitamin K2 (menaquinone). Vitamin K2 is recognized for its essential role in inhibiting calcium deposits in arteries and blood vessels. Supplementary vitamin K2 exists in several forms, but the most common are synthetic menaquinone-4 (MK-4) and natural menaquinone-7 (MK-7), where the latter is more effective.

 

Usage tips

1 to 2 tablets a day, orally, for a body weight of 70 kg or less, morning and/or evening, before or with meals. Duration of use varies according to individual needs: from a few weeks to several months.

 

 

Composition

Composition for 2 tablets: CSE, Castanea sativa fruit extract (34 mg), POE, Portulaca oleracea extract (34 mg), vitamin C (L-ascorbic acid; 40 mg or 50% of NRV*), vitamin K2 MK-7 (menaquinone; 75 μg or 100% of NRV*), Castanea sativa fruit flour, microcrystalline cellulose, hydrated silica, magnesium stearate.
